Determination of carboplatin in canine plasma by high‐performance liquid chromatography

Abstract: Carboplatin is an antineoplastic drug administered to treat different tumoral conditions in canine oncology. The objective of this study was to validate a high-performance chromatographic (HPLC) method which could be applied in canine pharmacokinetic studies. Following ultrafiltration using a Centrifree device, standards, quality controls and plasma samples were separated by isocratic reversed-phase HPLC on an Inertsil ODS-2 (250 x 4.6 mm i.d.) analytical column and quantified using UV detection at 220 nm. “…The 5-FU concentration was measured with a biochemistry method in the lymph nodes. In our study, we presented, for the first time, the distribution of cisplatin with an optical method and in combination with the blood and lymph node cisplatin concentration [25][26][27][28]103]. The current information highlights the fact that inhaled chemotherapy can target micrometastases throughout the lymphatic system and opens the way to using this treatment modality as a neoadjuvant and or adjuvant for early-stage treatment.…”

“…In addition, the following techniques were used to evaluate, in other lymph node specimens from the same patients, the existence of cisplatin within the lymph nodes [25][26][27]. First, the wet acid digestion procedure for digestion of biological tissues and liquids was used.…”

“…Platinum in biological fluids is currently determined by atomic or mass spectrometric techniques usually based on plasma or flame atomization. In this study a validated method for platinum determination which was based on the Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES) technique after suitable optimization was used [59,[63][64][65]. For this purpose a Perkin Elmer Optima 3100 XL axial viewing spectrometer was employed for all platinum measurements.…”

“…Few LC-UV methods were reported but all of them suffered from poor sensitivity (limit of quantification (LOQ) in the order of a few µg mL −1 ). [269][270][271] The introduction of an additional derivatisation step improved sensitivity (up to a factor of 100) with LC-UV methods. 272,273 Better LOQ's, between 2 and 25 ng mL −1 , were reached with LC-MS. [274][275][276][277][278] However, the technique of choice for the detection of platinum derivatives was undoubtedly inductively coupled plasma mass spectrometry (ICP-MS).…”

“…This is because this analytical technique requires a sample degradation step via photolysis digestion. 113,[288][289][290] For the separation step, different chromatographic supports were used, including reversed phase columns, 269,270,277,281,278 ion exchange columns, 275 HILIC support 285 or cellulose based columns for the enantiomer resolution of oxaliplatin. 291 CE was also reported as a promising technique for the separation of platinum derivatives.…”
