Determination of Creatine and Creatinine in Urine

Abstract: An improved method for the determination of creatine and creatinine in urine using the Jaffe reaction is described. Accurate results for the determination of creatinine depend upon carefully controlling the hydrogen ion concentration of the reaction mixture, the temperature, and the heating time. The authors conclude that many of the disputes regarding the optimum conditions for the determination of creatine in urine may be resolved in the light of the very marked effect of hydrogen ion concentration.

“…Fuller ton, Calif.. USA); serum alkaline phosphatase was determined spectrophotometrically (Zeiss Spcktralphotometer PM2D, FRG) accord ing to the method of Bessey [1946]; serum creatinine was determined by picric acid compound with a Zeiss spectrophotometer | Clark and Thompson, 1949]. and serum hydroxyproline was assayed spectrophotomctrically (Bausch & Lomb, FRG) with pyrrole and pdimethylbenzaldehydc [Stegemann, 1958;Woessner, 1961],…”

“…After the collection, urinary total noradrenaline and adrenaline excretions were determined by the method of Davidson and Fitzpatrick ( 25 ). Urinary creatinine excretion was measured by the method of Clark and Thompson ( 26 ). After being fed for 28 d [on day 29], the rats were anesthetized by intraperitoneally injecting of ␣ -chloralose and urethane (75 and 750 mg/kg of body weight, respectively).…”
