Determination of Docetaxel and Paclitaxel in Human Plasma by High-Performance Liquid Chromatography

Abstract: Taxanes, docetaxel and paclitaxel, represent important antineoplastic agents with broad spectra of antitumor activity. The authors developed and validated a high-performance liquid chromatography method with ultraviolet detection for quantifying both taxanes in human plasma. The assay uses liquid-liquid extraction as sample treatment and an isocratic mobile phase and reversed-phase chromatography to determine docetaxel with paclitaxel as internal standard and vice versa. The lower limit of quantification was 0… Show more

“…Numerous analytical methods for determination of taxanes in biological fluids have been proposed [9][10][11][12][13][14]. Most of them rely on HPLC separation with UV [9][10][11][12] or mass detection [13,14].…”

“…Therefore, there is a need for a simple and rapid method, which permits fast screening of samples in polymeric matrices. There are several reports of docetaxel being quantified in biological matrices using either HPLC-UV [19] [20] alone or HPLC coupled to mass spectrometer [21]- [30]. However, LC-MS/MS represents to be the preferred analytical instrument to quantitatively analyse drugs in various matrices due to the inherent specificity and sensitivity.…”

“…The structures and molecular weights of paclitaxel and docetaxel differ enough to eliminate the possibility of errant findings using LC-MS/MS. 22 Pre-existing surface contamination with docetaxel likely explains these findings. Surface swipe test monitoring and thorough cleaning of affected surfaces are necessary to reduce environmental exposure.…”
