Determination of Furazolidone and Nitrofurazone in Chicken Tissues

Abstract: F A method for the determination of furazolidone and nitrofurazone in chicken tissues is based on the formation and colorimetric estimation of 5nitro-2-furaldehyde phenylhydrazone. A chromatographic separation of the phenylhydrazone from nonspecific liver chromogens permits visual demonstration of the nitrofuran in concentrations as low as 0.5 p.p.m. and permits quantitative determination of these nitrofurans in chicken liver at 1 to 5 p.p.m. with an accuracy of 92 to 104%. A nonchromatographic method is used … Show more

“…T h e colorimetric methods of Buzard et al (1956) and Herrett 8c Buzard (1960) for estimating FZ are not specific as they d o not differentiate between the parent drug and its metabolites. However, the HPLC method of Ernst & Van der Kaaden (1980) used in this investigation is both specific and sensitive.…”

“…Using the colorimetric method of Herrett & Buzard (1960), with a limit of detection of 0.5-5 pg g-', no FZ concentrations could be detected in any of the tissues examined. However, using the HPLC method of Ernst & Van der Kaaden (1980) with a limit of detection of 0.05 pg g-', FZ was detected in the gluteal muscle and liver at concentrations of 0.26 f 0.01 and 0.10 k 0.02 pg g-I, respectively, 24 h after the end of the dosage period (10 mg/kg, 5 days).…”

“…portion of the toluene phase was mixed with 1 g. of aluminum oxide in a centrifuge tube. The 5-nitro-2-furfuraldehydephenylhydrazone adsorbed t o the alumina, showing a red color which was a qualitative test for that specific hydrazone (9). T o elute the hydrazone.…”
