Determination of medroxyprogesterone acetate residues by CE immunoassay with chemiluminescence detection

Abstract: A rapid and simple method is developed for the determination of medroxyprogesterone acetate (MPA) by CE immunoassay with chemiluminescence (CL). This method is based on the competitive reactions between horseradish peroxidase (HRP)-labeled MPA (MPA-HRP) and free MPA with anti-MPA antiserum. The influencing factors on the electrophoresis and CL detection were studied completely and the optimal conditions of separation and determination were obtained. The linear range was 2.0-50 nmol/L and the LOD for MPA was 0.… Show more

“…Basically, this CL reaction has been used in the determination of medroxyprogesterone acetate (MPA) [121] in pork tissues. MPA is a synthetic hormone often used in veterinary medicine to speed up fattening and can be a potential hazard for consumers.…”

“…In addition, medroxyprogesterone acetate (MPA), a tumor marker, carbohydrate antigen 15-3 (CA15-3), and a tumor Ag, CA 125 have been detected by the CE immunoassay coupled with a HRP label. [23][24][25] The sensitivity of CL makes it very promising for microchip CE-detection systems. Tsukagoshi et al 26 developed a micro total analysis system (m-TAS) incorporating the CL reaction of isoluminol isothiocyanato-microperoxidase-hydrogen peroxide.…”

“…It has been reported worldwide that some synthetic steroidal hormones have been used as growth promoters in animal breeding (Barel et al, 2006;Hooijerink, Van Bennekom, & Nielen, 2003;Peng, Xu, Jin, Chu, & Wang, 2006b;Peng, Huo, Liu, Chu, & Xu, 2007;Peng et al, 2008Peng et al, , 2009Xu, Peng, Hao, Jin, & Chu, 2006;Yuan, Xu, & Peng, 2008;Zhou, 2003). The potential harm of these hormone residues in foods and the environment has been recognised publicly.…”

“…MPA is also considered as a hormone which can speed up fattening. The presence of trace MPA in animal tissues poses a potential hazard for normal consumers Peng et al, 2007). The determination of MPA concentration in the samples of serum, urine and tissue is of great importance in daily life in China.…”
