Determination of Polynuclear Aromatic Hydrocarbons in Seafood by Liquid Chromatography with Fluorescence Detection

Abstract: Modification of a previously published method for determination of polynuclear aromatic hydrocarbons (PAHs) produces very clean seafood extracts in less than half the time. After alkaline digestion of the seafood, PAHs were partitioned into 1,1,2- trichlorotrifluoroethane. The resulting extract was cleaned up by solid-phase extraction on alumina, silica, and C18 adsorbents and then analyzed by gradient reversed-phase liquid chromatography with programmable fluorescence detection. Average recoveries of 12 PAHs … Show more

“…Over 100 PAHs have been identified and occur as complex mixtures, never as individual components. The two most common methods to measure PAHs are by HPLC with fluorescence detection (Perfetti et al, 1992;Dafflon et al, 1995;Chen et al, 1996) and gas chromatography with mass spectrometry detection (GC-MS) (Nyman et al, 1993;Chen and Lin, 1997;Chiu et al, 1997). Given the complexity of cooked food matrixes such as grilled meats, which contain numerous aromatic and heterocyclic aromatic compounds, in addition to PAHs, highly selective and specific methods are required for unequivocal analyte detection.…”

“…Apesar da pouca solubilidade em água, os mesmos podem ser transportados em meios aquáticos, adsorvidos em partículas em suspensão, ficando posteriormente, depositados nos sedimentos 11 . Devido a habilidade em filtração de água e por não apresentarem capacidade de biotransformá-los, determinados animais marinhos, como ostras e mexilhões, acabam acumulando os HPAs em seus organismos 12 .…”

“…These compounds are ubiquitous environmental pollutants that are of concern because of their mutagenicity and/or carcinogenicity. [1][2][3] Intensive investigations have been carried out on the determination of toxic organic compounds emitted into the atmosphere because of the progressive environment degradation. In plant samples and food, the presence of PAHs is attribute exclusively to environment contamination.…”
