Determination of salbutamol in human plasma and urine using liquid chromatography coupled to tandem mass spectrometry and its pharmacokinetic study

Abstract: A sensitive and selective liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) was developed and validated for the determination of salbutamol in human plasma and urine, and successfully applied to the pharmacokinetic study of salbutamol in Chinese healthy volunteers after inhalation of salbutamol sulfate aerosol. Salbutamol and the internal standard (IS) acetaminophen in plasma and urine were extracted with ethyl acetate, separated on a C(18) reversed-phase column, eluted with mobile phase of … Show more

“…According to the literature, after oral administration of salbutamol to ten healthy volunteers, the urinary excretions of unchanged drug and sulfate conjugates were 31.8 ± 1.9% and 48.2 ± 7.3%, respectively . Furthermore, the concentration of salbutamol in plasma was reported to be much lower than that in urine after the oral administration of salbutamol . Thus, much more metabolites of (R)‐salbutamol in urine were detected than those in plasma or feces.…”

“…The clinical use of salbutamol is the management of reversible airways observation such that occurs in asthma and delaying premature labour. 1 Literature survey shows that several spectrophotometric, 2-12 spectrouorimetric, 13 conductometric, 14 potentiometric, 15 LC-MS 16 and HPLC [17][18][19][20][21][22][23] methods reported for the determination of salbutamol sulfate in various samples. So far, only one kinetic spectrophotometric method has been reported for determination of salbutamol sulfate.…”

“…6 Various techniques and analytical methods have been developed for the measurement of SAL residues in various matrices, including feedstuffs, animal tissues, urine, and so on. 5,[7][8][9][10][11][12][13] The preferred and official analytical methods for SAL are high-performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS), with LODs of 18 ng mL À1 and 1 ng mL À1 . However, these instrumental analysis methods usually require the steps of sample preparation and analysis, and are of high cost and relatively time-consuming.…”
