Determination of SARS‐coronavirus by a microfluidic chip system

Abstract: We have developed a new experimental system based on a microfluidic chip to determine severe acute respiratory syndrome coronavirus (SARS-CoV). The system includes a laser-induced fluorescence microfluidic chip analyzer, a glass microchip for both polymerase chain reaction (PCR) and capillary electrophoresis, a chip thermal cycler based on dual Peltier thermoelectric elements, a reverse transcription-polymerase chain reaction (RT-PCR) SARS diagnostic kit, and a DNA electrophoretic sizing kit. The system allows… Show more

“…Holes were punched out of the PDMS slab to form reservoirs for introduction of liquid. The bottom layer was fabricated by wet etching method as described previously [21,22], which included 20 microcolumns. Each microcolumn is 5 mm high, 150 mm wide and 1 mm long.…”

“…For example, chip electrophoresis of viral RNA, or DNA produced via PCR, served for genotyping Hepatitis B [1] and Hepatitis C virus [2], for the detection of Dengue-2 virus [3], of the Coronavirus causing severe acute respiratory syndrome (SARS) [4], for analysis of DNA samples of Cytomegalovirus [5] and Herpes simplex virus [6]. Only in a few cases, the viruses themselves were subjected to electrophoresis; e.g.…”
