Background: Solifenacin succinate is a competitive muscarinic acetylcholine receptor antagonist used in the treatment of overactive bladder with or without urge incontinence. Methods: Liquid chromatography-tandem mass spectrometry method was used for quantification of Solifenacin (SF) in rat plasma. Solifenacin-d5 (SFD5) used as an internal standard. Chromatographic separation was performed Gemini-NX C18, 50 × 4. Conclusions: This method is successfully applied in the Pharmacokinetic study of rat plasma.

Four sensitive, precise and accurate spectrophotometric methods for the estimation of solifenacin succinate (SFS) have been developed. Method A describes the interaction of SFS with potassium permanganate in alkaline medium to give green colored manganate ion with absorption maxima at 610 nm. Methods B and C are based on the formation of ternary complexes of SFS with, copper (II)/eosin (method B) and ammonium molybdate/ammonium thiocyanate (method C), respectively which are extracted into chloroform and have absorption maxima at 545 nm (method B) and 465 nm (method C). Method D was based on the formation of yellow colored ion-pair complex between bromocresol green and SFS in dichloromethane medium with absorption maxima at 415 nm. Regression analysis of Beer's law plot showed good correlation in the concentration range of 5-50, 2.5-50, 10-100 and 2-20 μg/mL for methods A, B, C and D, respectively. Different variables affecting the reaction were studied and optimized. The proposed methods were applied successfully for the analysis of SFS in tablets dosage forms. No interference was observed from common pharmaceutical excipients.

Enantioseparation of 1-Phenyl-1,2,3,4-tetrahydroisoquinoline on Polysaccharide-Based Chiral Stationary Phases

The enantiomers of 1-phenyl-1,2,3,4-tetrahydroisoquinoline have been directly separated on polysaccharide-based chiral stationary phases (CSPs). The normal phase separation of (S)-and (R)-1-phenyl-1,2,3,4-tetrahydroisoquinoline was accomplished by screening of the immobilized Chiralpak IC column with different eluents. The effect of mobile phase type on retention, selectivity and resolution was studied. 2-Propanol or ethanol/n-hexane/ ethanolamine mixtures were applied as mobile phases by screening of following polysaccharide-based immobilized (Chiralpak IA, Chiralpak IC) and coated (Lux Cellulose-1, Lux Cellulose-2, Lux Amylose-2) CSPs. Polar organic and reversed-phase conditions were also tested for direct enantioseparation of 1-phenyl-1,2,3,4-tetrahydroisoquinoline.
