Development and characterization of specific anti‐Usutu virus chicken‐derived single chain variable fragment antibodies

Abstract: Usutu virus belongs to the Japanese encephalitis serogroup within the Flaviviridae family. Mammals may become incidental hosts after the bite of an infected mosquito while birds act as the main reservoir. Human cases have become more common recently and elicit various outcomes ranging from asymptomatic to severe illness including encephalitis. Problematically, antisera against Usutu virus cross-react with other flaviviruses such as the cocirculating West Nile virus. As an approach to generate Usutu virus-speci… Show more

“…The survival rate against JEV with 1b-4/21-C12 treatment is relatively low compared to the 70% in mice inoculated with phage display peptide or JEV E stem-derived peptide [13,14]. The efficacy of our peptide might be enhanced on combination with ribavirin, scFv, and FDA-approved drugs [20][21][22]. Future studies should measure the half-lives of peptides conjugated with various fatty acids and determine the exact mechanism of action of 1b-4/21-C.…”

“…The number is close to the library size of our present study (3.1 × 10 8 CFU/ml) 56 . In 2020, Schoenenwald et al developed four chicken-derived anti-Usutu virus scFvs 57 . Moreover, Ge et al produced IgY-derived scFv from chicken species that target the Canine Parvovirus capsid protein 58 .…”

“…Moreover, Ge et al produced IgY-derived scFv from chicken species that target the Canine Parvovirus capsid protein 58 . In both investigations, about 100 clones were selected for the first time during the screening procedure to capture the best binders 57 , 58 . The number of clones is close to our number of screening clones (120 clones).…”

“…Compared with IgG, it was easy to prepare and can be mass-produced by immunizing laying hens ( 25 , 26 ). What's more, it did not react with rheumatoid factor, mammalian Fc receptor, and complement ( 27 ), and had barely cross-reaction with IgG ( 28 ), showing a high specificity. As a result, IgY has a broad application prospect in the development of biological products and the prevention and treatment of diseases.…”
