Development and evaluation of novel recombinant adenovirus-based vaccine candidates for infectious bronchitis virus and Mycoplasma gallisepticum in chickens

Abstract: Avian infectious bronchitis caused by the infectious bronchitis virus (IBV), and mycoplasmosis caused by Mycoplasma gallisepticum (MG) are two major respiratory diseases in chickens that have resulted in severe economic losses in the poultry industry. We constructed a recombinant adenovirus that simultaneously expresses the S1 spike glycoprotein of IBV and the TM-1 protein of MG (pBH-S1-TM-1-EGFP). For comparison, we constructed two recombinant adenoviruses (pBH-S1-EGFP and pBH-TM-1-EGFP) that express either t… Show more

“…A recombinant adenovirus was constructed containing the TM-1 protein of MG (pBH-S1-TM-1-EGFP) and S1 spike glycoprotein of infectious bronchitis virus. The recombinant bivalent pBH-S1-TM-1-EGFP adenovirus construct vaccine provides potential protective effects against bronchitis and MG infection ( Zhang et al., 2018 ). Therefore, preventive measures against MG infection and other respiratory pathogens must take into account by vaccination and/or antimicrobial agents.…”

“…Nonpathogenic and attenuated bacteria may be used as vectors, but nonpathogenic species are preferred ( Liljeqvist and Stahl, 1999 ). The development of recombinant vaccines by cloning and identification of important MG surface antigens and colonization factor, development of expression and transformation strategies bewitch the interest of scientist ( Zhang et al., 2010 , 2018 ; Shil et al., 2011 ). Gene transfer is accomplished in mycoplasma species by electroporation ( Liu et al., 2000 ; Pour-El et al., 2002 ; Mudahi-Orenstein et al., 2003 ) and Enterococcus faecalis –mediated conjugation ( Ruffin et al., 2002 ).…”

“…Numerous attempts have been made for vaccine development against Mycoplasma microorganisms, exploring DNA, proteins, or capsular polysaccharides for effective control. Novel approaches include recombinant protein or DNA vaccines [117,118], subunit vaccines [119,120], nanoparticle-based vaccines [121,122], plant-based or edible vaccines [123,124], and cytokine or TLR-based vaccines [125,126,127].…”

“…Hence, it can also be investigated as a vaccine candidate. Recombinant vaccine constructs using the TM-1 protein of Mycoplasma gallisepticum (pBH-S1-TM-1-EGFP) have been studied by Zhang et al [118].…”

“…IBV belongs to the genus Coronavirus, family Coronaviridae, of the order Nidovirales (Mayo and Pringle 1998). The IBV genome consists of a single-stranded positive-sense RNA that encodes four structural proteins [spike (S), membrane (M), envelope (E), and nucleoprotein (N)] (Feng et al 2017;Zhang et al 2018Zhang et al , 2019. IBV primarily infects the respiratory tract, kidneys, and reproductive system of poultry, which can cause difficulty breathing and an enlargement of the kidneys (Cavanagh 2007).…”
