Development and validation of short-amplicon length PCR assay for macaques meat detection under complex matrices

Abstract: The macaque (Macaca fascicularis) monkeys are the third largest primate population in tropical forests. Being the potential carrier of Simian Immunodeficiency and Ebola and Corona viruses, as well as being a religiously-protected and wildlife-protected species could not save the macaques from being over-hunted and consumed. Despite having needs, methods to detect monkey species in foods are rarely documented. To fill this gap, this article describes a monkey-specific polymerase chain reaction assay targeting a… Show more

“…Results of this study suggest that the value allocated to baboons by park-adjacent dwellers is related mainly to consumptive uses. Primate meat consumption has been reported previously in many countries (Ali et al 2017;Blair et al 2017;De Thoisy et al 2005;Shaffer et al 2018;Tshikung et al 2019). In southeastern DRC for example, identified a notable quantity of baboon carcasses from parks and natural reserves illegally sold in urban markets of Lubumbashi, a city located nearly 150 km from the study area.…”

“…The acceptability and value of buffalo meat vary culturally and geographically. In Europe and China, where it is widely accepted and eaten, it has a higher value than beef and is considered a delicacy (Hossain, Ali, Hamid, Hossain, et al, 2017;Wang, Hang, et al, 2019). In India, however, buffalo meat is less valued and is considered an adulterant of more valuable meat such as mutton (Naveena et al, 2017).…”

“…Products from PCR amplifications are subjected to enzymatic restriction (cutting) by specific endonucleases, resulting in a specific fingerprint in the product that is then used to identify the target species. Publicly available web domains provide online tools for researchers to perform in silico analyses to determine species-specific endonuclease and endonuclease sites for their targets (Ali et al, 2017). To detect Malaysian box turtle in food and traditional Chinese medicines, Asing et al (2016b) used the PCR-RFLP technique to restrict the 120 base pair (bp) long product of the cyt b gene with the Bfa1 endonuclease.…”

“…There is also a concern about PCR product carryover contamination due to the 10 8 -fold amplification of the target [26]. Finally, shorter DNA targets would be thermodynamically more stable than longer ones [27] and choosing a shorter target is favourable since it can withstand decomposition states [28].…”
