Development of a Colloidal Gold-based Immunochromatographic Test Strip for Screening of Microalbuminuria

Abstract: A rapid immunochromatography (ICG) assay based on antibody colloidal gold nanoparticles specific to human serum albumin (HSA) was developed, and its applications for primary screening of HSA in the urine were evaluated. A monoclonal antibody (MAb) specific to HSA was produced from cloned hybridoma cells (EMRC1) and used to develop an ICG strip. The nanocolloidal gold, with an average particle diameter of 20 nm, was synthesized and labeled MAb as the detection reagent. An antibody colloidal gold probe was appli… Show more

“…Following the blockade of the non-specific binding sites, the wells were treated with antiinsulin mAb (monoclonal antibody) (1 μg/well), and incubated at 37°C for 1 h. The residual binding was detected with anti-mouse HRP (horseradish peroxidase) conjugated antibody and compared with the standard curve. The amount of insulin was determined from a calibration curve based on the known concentrations of insulin [33][34][35][36].…”

“…Following the blockade of the non-specific binding sites, the wells were treated with the anti-insulin specific antibody at a concentration of 1 μg/well. The residual binding was determined by adding the peroxidase-labeled anti-mouse IgG secondary antibody and expressed as a percentage of the optical density value given by a control [34,37].…”

“…The use of immunochromatographic strips (ICS) has becoming widespread for the rapid and on-site detection of toxins, heavy metals, proteins, pesticides, hormones, drugs and pathogens (Wang et al, 2005Huo et al, 2006;Li et al, 2009;Omidfar et al, 2011;Yang et al, 2011;Liu et al, 2012). However, few colloidal gold-based immunochromatographic assay techniques have been published for the detection of aquatic animal pathogens.…”

“…Immunoglobulins can be directly attached on the surface of GNPs by the non-covalent reactions during the conjugation (Omidfar et al, 2010). The efficacy of the conjugate is strongly associated with the size and quality of the colloidal gold NPs (Gui et al, 2008;Omidfar et al, 2011).…”
