Development of a Novel Fluorophore for Real-Time Biomonitoring System

Abstract: Rapid in-field diagnosis is very important to prevent the outbreak of various infectious and contagious diseases. Highly sensitive and quantitative detection of diseases can be performed using fluorescent immunochemical assay with specific antigen-antibody binding and a good quality fluorophore. This can lead to the development of a small, portable, quantitative biosensor to transmit diagnostic results to a control center in order to systematically prevent disease outbreaks. In this study, we developed a novel… Show more

“…The fluorescent emission spectra also exhibited wide spectral ranges, and therefore, the presence of fluorescent latex beads could be easily detected with the emission optical filter (590 nm wavelength) as employed in the LED-based portable strip reader. This result was also consistent with the previous report of coumarin-derived dendrimer characterization 28 .…”

“…Coumarin-derived dendrimer was provided by Professor Hak-Sung Kim (Wonkwang University, Korea) and characterized previously 28 . For covalent conjugation of mAb with latex and coumarin-derived dendrimer, 0.8 mL of aliphatic amine latex beads (20 nm) were washed with PBS (pH 7.4) and in the presence of 0.8 mL conjugation buffer (0.1 M sodium bicarbonate, pH 8.5), 128 μL of coumarin-derived dendrimer (1 mg/mL in DMSO) was added to latex in the conjugation buffer at RT for 1 hour with shaking.…”

“…As shown in Figure 6, the 14 studies incorporating qualitative methods use open-ended questionnaire items, focus groups, and/or participant interviews to investigate qualitative questions relating to treatment outcomes, 29,84,89,105,123 treatment choices, 115 patient experiences, 86,110,122 and protocol compliance. 105 One study also engages participant observation to document outcomes.…”

“…The coumarin-derived dendrimer-based fluorescence-linked immunosorbent assay (FLISA) could detect two specific antigens of malaria: Histidine-rich protein II (HRP2) and lactate dehydrogenase (LDH). FLISA has good spectroscopic properties, such as photostability [ 146 ], and a better performance than traditional ELISA, enabling the quantification of the number of parasites in a sample even if they are present in low concentrations. Accordingly, FLISA method could be useful to detect asymptomatic cases at a modest price and with a high capacity.…”
