Development of an automated plant culture system

Abstract: An apparatus was constructed that could be used to grow plant tissues, organs, and whole plantlets under sterile conditions. This system accommodated independent or multiple concurrent growth of cultures. Growth of plants either equalled or exceeded that observed using the manual transfer procedure. The automated plant culture system (APCS) consists of silicone tubing, 2 impeller pumps, 2 glass medium reservoir bottles, a 3-way stainless steel vane, a plant culture chamber, and an interface module containing r… Show more

“…Similar to the current observation the positive influence of liquid medium on fresh mass development was reported for African oil palm (Teixeira et al, 1995;Kanchanapoom and Chourykaew, 1998) and date palm (Tisserat and Vandercook, 1985;Al-Khayri, 2012). A higher mass of African oil palm callus as compared to solidified medium was also reported using TIS (Sumaryono et al, 2008).…”

“…Adding BAP to the cell suspension cultures seemed to be controversial. Using liquid medium with NAA and without cytokinins as we found, was reported to be optimal in some date palm liquid cultures (Tisserat and Vandercook, 1985;Sharma et al, 1986;Saker et al, 2007). In contrast to our results, Gadallah (2007) reported higher fresh mass and somatic embryo number in date palm cv.…”

“…Taking into account the liquid condition of the medium, a system was initially designed that consisted of a large, elevated culture chamber that could be drained and then refilled with fresh medium (Tisserat and Vandercook, 1985). Subsequently a semi-automated system was developed in which plants were cultured in a large container on a medium that contained no gelling agent, with automatic addition and removal of liquid medium at fixed intervals so the plant material entered into contact with the medium several times throughout the day (Aitken-Christie and Davies, 1988).…”
