Development of an enzyme-linked immunosorbent assay for the determination of florfenicol and florfenicol amine in eggs

Abstract: A monoclonal antibody against florfenicol amine (FFA) was produced, and a competitive enzyme-linked immunosorbent assay (ELISA) was developed for detecting florfenicol (FF) and FFA in eggs. This proposed method exhibited the 50% inhibiting concentrations (IC 50 ) of 0.120 ng/mL for FF and 0.118 ng/mL for FFA under the optimization of experimental parameters. At the fortified levels of 0.4, 0.8, 2.4, and 30.0 μg/kg, the inter-assay recoveries of the target compounds ranged from 78.3 to 103.2%, and the intra-ass… Show more

“…MAb production consists of three steps: mouse immunization, cell fusion, and ascites fluid purification. 26,27 First, five female mice (6-weeks old) were immunized five times (a dose of 100 μg at the first immunization and 50 μg at booster immunization) by subcutaneous injection using CDM-S 1 -BSA, with an interval of three weeks. 28 The positive mouse with high affinity and sensitivity was screened by ic-ELISA, and immunized by intraperitoneal injection (25 μg) after the last booster immunization.…”

“…24,25 Immunoassays are alternative methods for the screening of various hazardous substances in the environment and foods. [26][27][28][29] In the case of anthranilic diamide insecticides, Zhang et al prepared a monoclonal antibody (mAb) and developed an indirect competitive ELISA (ic-ELISA) for the rapid detection of CYA. 30 The ic-ELISA provided a detection range of 0.43-6.15 μg L −1 and good recovery in pakchoi and tap water samples.…”
