Development of an ImageJ-based method for analysing the developing zebrafish vasculature

Abstract: Zebrafish with fluorescently labelled blood vessels provide an excellent model for studying angiogenesis. Most commonly the growth of the intersegmental blood vessels is investigated in response to compounds or manipulation of gene expression and analysed using manual methods, typically scoring the connectivity of these blood vessels to the dorsal longitudinal anastomotic vessel. Such methods are laborious and best suited to time points after the connectivity of these vessels has been established. By contrast,… Show more

“…Images and movies of non-injected and injected (vehicle and PGE2) zebrafish embryos were recorded using a stereoscopic zoom microscope (Nikon SMZ1500) and a fluorescence microscope (Olympus MVX10). Images were further processed and quantified using ImageJ (Simms et al, 2017) and ZFIN (Chávez et al, 2016) software. Movie files were converted and processed using Wondershare Video Converter.…”

“…To that end, HUVECs were encapsulated in BoneMA hydrogels precursors (5% (w/v)) containing LAP (0.15% (w/v)) at a cell density of 1 x 10 7 cells/ml, crosslinked for 15, 30 and 45 s as described previously. Vascular capillary network formation in these hydrogels was characterized daily for 7 days and quantified for the number of segments, number of end points and junctions, as well as the total length of the network using an ImageJ, as described previously [21].…”

“…Five embryos in each group were randomly selected and scanned. The zebrafish vasculature formation of DLAV and ISV after 20 h treatment with compounds was quantified using ImageJ as previously described [38]. The experiments were repeated twice with similar results.…”
