Development of an Ultrahigh-Performance Liquid Chromatography–Electrospray Ionization–Tandem Mass Spectrometry Method for the Simultaneous Determination of Salicylic Acid, Jasmonic Acid, and Abscisic Acid in Rose Leaves

Abstract: This paper describes a method to detect and quantitate the endogenous plant hormones (±)-2-cis-4-trans-abscisic acid, (−)-jasmonic acid, and salicylic acid by means of ultrahigh-performance liquid chromatography−tandem mass spectrometry (UHPLC−MS/MS) in hybrid rose leaf matrices. Deuterium-labeled [ 2 H 6 ] (+)-2-cis-4-trans-abscisic acid, [ 2 H 6 ] (±)-jasmonic acid, and [ 2 H 4 ]-salicylic acid were used as internal standards. Rose samples (10 mg) were extracted with methanol/water/acetic acid (10:89:1) and … Show more

“…The LC-MS/MS method was linear for all analytes over the concentration range (10-500 ng mL -1 for Cy, Dp, Pg, Pt; and 2-100 ng mL -1 for Mv, Pn, Cy3Glu, Cy3Gal, Cy3Ara, Dp3Gal, Mv3Gal, Pg3Glu, Pt3Glu) studied in both plasma and urine. The resultant r 2 values for all anthocyanidins and anthocyanins were close to 1 (Table 1), which indicated good linearity within the considered concentration ranges (Bosco, Daeseleire, Van Pamel, Scariot, & Leus, 2014). The LOD and LOQ values of all anthocyanidin and anthocyanin analytes were calculated in blank plasma and urine to determine the sensitivity of the method.…”

“…Then, 1.0 g of the homogenized sample was immersed in a 10.0-mL mixture of methanol-water-acetic acid (10:89:1, v/v/v) for 16 h at 4 C in the dark to perform extraction. 24 After 10 min of centrifugation at 4000 rpm, the supernatant was filtered through a 0.22-μm membrane (Xingya Scavenging Material Company, Shanghai, China). Then, the filtrate was rotary evaporated to yield a residue that was subsequently reconstituted in 250 mL ultrapure water and stored at 4 C prior to analysis.…”

“…The measurement of endogenous hormonal levels in plants has received a great deal of interest owing to their potential role in signaling networks and molecular mechanisms. Consequently, several analytical methods have been adapted to measure plant hormones, including enzyme-linked immunosorbent assay (ELISA), high-performance liquid chromatography with diode-array and fluorometric detection (HPLC-DAD/FLD), liquid chromatography-mass spectrometry (LC-MS), and gas chromatography-mass spectrometry (GC-MS) [ 10 , 11 ]. Among these, mass spectrometry (MS) methods have been found to provide high sensitivity and selectivity for analysis of plant hormones [ 12 ].…”

“…Among these, mass spectrometry (MS) methods have been found to provide high sensitivity and selectivity for analysis of plant hormones [ 12 ]. GC-MS is routinely applied for the simultaneous analysis of various hormones in plant tissues; however, it is relatively time consuming when compared to LC-MS [ 11 ]. Although several LC-MS methods have been developed for simultaneous analysis of multiple plant hormones [ 11 15 ], there is no any single suitable LC-MS method for simultaneous analysis of BR and other major hormones involved in drought stress.…”
