Development of droplet digital PCR for the detection of Tilletia laevis, which causes common bunt of wheat, based on the SCAR marker derived from ISSR and real-time PCR

Abstract: Common bunt of wheat caused by Tilletia laevis and/or T. caries (syn. T. tritici), is a major disease in wheat-growing regions worldwide that could lead to 80% or even total loss of production. Even though T. laevis can be distinguished from T. caries on the bases of morphology of teliospores using microscopy technique. However, molecular methods could serve as an additional method to quantify the pathogen. To develop a rapid diagnostic and quantify method, we employed the ISSR molecular marker for T. laevis i… Show more

“…As there is an increasing interest in the ecological and evolutionary dynamics in tripartite systems in microbial communities such as hosts, helper viruses and satellite viruses, the presented methods for multiplex ddPCR should be considered for other systems as well. This method can help the study of the role of virophage-giant virus interactions, and has the potential to be used for a wide range of disciplines, for clinical samples to identify drug-resistant subpopulations [ 36 ], agricultural evaluation of pest resistance [ 37 ], and field surveying of viruses [ 29 ] among others.…”

“…The grains were sterilized and vernalized, and ten wheat seedlings were transplanted into every pot. Based on morphological characterization of teliospores of T. laevis ( Supplementary Figure 1 ) and specific band and sequence of SCAR marker of T. laevis ( Supplementary Figure 2 ), we identified the pathogen of T. laevis based on the previous reports ( Xu et al, 2020 ; Qin et al, 2021 ). Then, the identified T. laevis was use in this study.…”

“…A droplet digital PCR (ddPCR) sensing system for detecting common bunt disease (caused by Tilletia laevis) in wheat was reported in [42]. The ddPCR exhibited superior detection limit of 30 fg/µL as compared to qPCR (100 fg/µL).…”
