Tobacco ringspot virus (TRSV) is a plant quarantine virus in Korea. As such, a TRSV examination is conducted when importing various crops. In this study, RT-PCR and nested PCR systems for TRSV detection in quarantine sites, and the modified-positive control plasmid for proving laboratory contamination and false positive reactions were developed. The developed diagnostic system was used to detect TRSV in the quarantine site. It revealed that from 2012 to August 2014, a total of 12 cases were detected in imported various crops. The system is expected to continue contributing to TRSV detection in plant quarantine.

: Andean potato latent virus (APLV) is a phytopathogenic virus that belongs to the Group IV (+) sense ssRNA viruses of the genus Tymovirus. It mainly infects potatoes and is specified as a controlled quarantine virus in Korea. In this study, two primer sets of RT-PCR and nested PCR [set 2 (404 → 259 bp) and set 23 (501 → 349 bp)], were selected, which can rapidly and accurately diagnose APLV in quarantine sites. In addition, a modified-positive control plasmid is development, can possible verification of laboratory contamination in diagnosis of APLV detection. The PCR-base system developed in this study is expected to diagnose APLV and contribute to the plant quarantine in Korea.

The Cucurbitaceae are a plant family that consist of over a hundred genera, the most important of which are squash, pumpkin, zucchini, some gourds, cucumber and watermelon. These are among the top imported seeds in Korea. At the time of their import, the Squash mosaic virus (SqMV), the Cucumber green mottle mosaic virus (CGMMV) and the Kyuri green mottle mosaic virus (KGMMV) are designated as regulated viruses for quarantine in Korea. This study was conducted to develop specific primer sets for easy and rapid detection of SqMV, CGMMV and KGMMV. RT-PCR with the nested PCR primer sets and modified positive control plasmids were capable of highly sensitive detection and verification of such viruses. In addition, developed diagnostic PCR systems applied to quarantine sites detected 47 cases of SqMV, 67 cases of CGMMV and 17 cases of KGMMV between 2010 and the first half of 2014.
