Development of Tizanidine HCl-Meloxicam loaded mucoadhesive buccal films: In-vitro and in-vivo evaluation

Abstract: The purpose of the study was to develop Tizanidine HCl (TZN) and Meloxicam (MLX) loaded bilayer mucoadhesive films intended for buccal administration, aiming to enhance the bioavailability. Bilayer films were prepared by solvent evaporation technique selecting arabinoxylan (ARX) as a sustained release (SR) layer forming polymer and hydroxypropyl methylcellulose (HPMC) E-15 as an immediate release (IR) layer-forming polymer. Prepared films were subjected to in-vitro drug release, surface morphology, mechanical … Show more

“…Film-forming polymers, in the presence of other additives (e.g., plasticizers, penetration enhancer, flavors and sweeteners), play a critical role in the development of effective oral films. Previously, hydroxypropyl methylcellulose (HPMC-K15M) was investigated for the development of oral films [ 23 , 24 ]. The main reason for selecting HPMC is that it provides appropriate mechanical strength to the films [ 25 , 26 , 27 ].…”

“…2,8 Bilayer films are the drug delivery system that contains more than one active pharmaceutical agent in a double layered polymeric film. 9 Recently, bilayer films are gaining preference over single layer films as they offer a combination of unique properties of all substrates, which is often advantageous. 10 Bilayer films can be made as mucoadhesive films by using mucoadhesive polymers with the added advantage of incorporating a sustained drug release layer 11 and an immediate drug release.…”

“…Tizanidine hydrochloride (TZN) is a centrally acting α2-adrenergic agonist that acts as a muscle relaxant for the treatment of muscle spasms, cramping, and tightness caused by medical problems such as spine or central nervous system. 8 In addition, it has a high affinity for α2-agonist receptors, which reduces the nervous reflex and thus has an analgesic effect. Therefore, TZN is also prescribed off label for some symptoms of migraine and fibromyalgia.…”
