Dextran‐magnetite particles: Contrast‐enhanced MRI of blood–brain barrier disruption in a rat model

Abstract: Dextran-magnetite particles (DMP) were studied for their use as a MR contrast agent to visualize lesions with a blood-brain barrier (bbb) disruption. A freezing injury to the rat cerebral cortex was used as a model of bbb disruption. The biodistribution of iv-injected DMP was studied using atomic absorption spectrophotometry, electron microscopy, and MRI. One hour after injection, focal accumulation of the particles in capillary endothelial cells could be demonstrated in the freezing lesion. Despite the observ… Show more

“…In this study ultra-small particles of iron oxide (USPIO) were chosen because they have a very strong reducing effect on the signal intensity in T 2 -weighted imaging, which might help to find minor permeability changes in the pre-paretic period. An additional feature of USPIO is that they can be internalized by cells with macrophagic activity [22][23][24], such as macrophages, microglial cells, and also tumor cells and endothelial cells [25,26]. This proved to be a valuable tool to detect inflammatory areas [23,24].…”

“…Immediately the suspension was brought to 4OC and SV-magnetoliposomes were separated from uncomplexed SV andlor unencapsulated DMP by washing four times by centrifugation at 15000 x g for 5 min. The final pellet containing the magnetoliposomes was resuspended in BSS at 5 mg lipid/ml, stored at 4OC in dialysis tubing, and used for the labeling of cells within 48 h. The DMP-concentration of magnetoliposomes was assessed by graphite-furnace atomic absorption spectrophotometry as described for the assessment of DMP in plasma samples (27).…”

“…Brain edema induced by cold injury (cold lesion, cryogenic cortical lesion, freeze-traumatized lesion) is a well-known experimental model simulating trau matic brain edema, i.e., vasogenic brain edema, 1,5,14) and has been studied extensively. The disruption of the blood-brain barrier (BBB) associated with vaso genic edema has been extensively investigated since BBB disruption is considered to be important in the development of vasogenic brain edema.…”
