Diagnostic and Prognostic Relevance of Autoantibodies in Uranium Miners

Abstract: Uranium miners exposed to silica dust have a higher risk of developing systemic sclerosis (SSc) and systemic lupus erythematosus (SLE). Sera of 1976 former uranium miners were analysed for autoantibodies typical of connective tissue disease. The frequency of some of these antibodies (anti-centromere, -topoisomerase I, -nucleolar, -dsDNA, -Ro/SSA, -La-SSB and U1-RNP antibodies) was significantly higher compared to a gender- and age-matched control group and was associated with the intensity of exposure as well … Show more

“…Four of these cases developed CTD that was specific for the corresponding autoantibody, such as anti-RNP for MCTD or anti-SSA for Sjogren's syndrome, and two patients with positive MPO-ANCA developed vasculitis (microscopic polyangiitis). In contrast to ANA (24)(25)(26), only a few previous studies have reported the prevalence of specific autoantibodies in non-CTD individuals; anti-Ro, 2.7%-10.2% (33,34), anti-La, 0.9% (34), anti-Scl70, 0%-3% (35,36), anti-Sm, 0%-0.5% (34,35), which are similar to our results.…”

“…The type and prevalence of disease-specific ANAs have been investigated among specific cohorts, such as consecutive patients and uranium miners [19,20], but few largescale surveys have been conducted within the general population. In our study, the detection of disease-specific ANAs in serum samples from 2181 individuals yielded an anti-SSA/Ro prevalence of 2.7% (3.5% in females; 1.0% in males).…”

“…Patients with silicosis and other individuals highly exposed to silica have been shown to have increased autoantibodies (8,9), immunoglobulins, and immune complexes (10). However, a recent study of lupusprone mice demonstrated a decrease in serum immunoglobulin levels associated with the development of autoimmunity (11), and decreased levels of immunoglobulins were observed in foundry and pottery workers working in their jobs for an average of ϳ15 years (12).…”
