Dietary flavones suppresses IgE and Th2 cytokines in OVA-immunized BALB/c mice

Abstract: The results suggest that the diet containing one of the two flavones might suppress the up-regulation of serum IgE induced by OVA-immunization through the suppression of Th2-type immune response.

“…As IL-4 is the major inducer for class-switching to IgE biosynthesis in B lymphocytes, the decrease in serum IgE in cocoa-fed rats should be associated with lower IL-4 production. It is noteworthy that although some authors reported low levels of IL-4 after flavonoid treatment [30,31] others did not find any difference [12]. On the other hand, previous studies with a similar cocoa diet in rats noted a decrease of IL-4 production in spleen [16,18] but not in MLN cells [18].…”

“…In first case, a leaf extract rich in the flavonol myricitrin decreased total IgE in an allergy model in transgenic mice [29]. The flavones chrysin and apigenin suppressed total IgE concentrations whereas IgG, IgM and IgA concentrations were not affected [30] but later studies show that apigenin in a mice model of atopic dermatitis decreased serum total IgG1 and IgE concentrations, without modifying significantly specific antibodies, and ameliorated the development of skin lesions [12].…”

“…The mice received OVA (100 lg body -1 ) with an alum adjuvant (LSL Inc., Japan) by intraperitoneal (i.p.) injection on days 7 and 21 to elicit a specific IgE response (Yano et al 2007). After 5 days of the second OVA induction (day 26), blood and splenocytes were collected.…”

“…It was also found that nobiletin, a polymethoxyflavonoid, when given intraperitoneally to OVA-sensitized rats at a dose of 1.5 or 5 mg/kg, reduced OVA-induced increases in eosinophils and eotaxin expression (Wu et al, 2006). In subsequent investigations, flavonoids such as quercetin, isoquercitrin, rutin, 3-O-methylquercetin 5,7,3',4'-O-tetraacetate, narirutin, apigenin, luteolin, sulfuretin, hesperdin, fisetin and kaempferol have been shown to suppress responses in various types of allergic animals (Makino et al, 2001;Fernandez et al, 2005;Rogerio et al, 2007;Jung et al, 2007;Jiang et al, 2007;Funaguchi et al, 2007;Yano et al, 2007;Cruz et al, 2008;Choi et al, 2009;Li et al, 2010;Leemans et al, 2010;Shishebor et al, 2010;Kim et al, 2011;Wu et al, 2011;Gong et al, 2012).…”
