Differential accumulation of a protein kinase homolog in Trypanosoma brucei

Abstract: Using degenerate oligonucleotide primers derived from conserved regions in the catalytic domains of protein kinases, we have identified transcripts of the protein kinase families in Trypanosoma brucei by the polymerase chain reaction technique. From the cDNAs synthesized from poly(A)+ RNA purified from the bloodstream form of the pathogen, we have obtained seven distinct partial cDNA sequences. Deduced amino acid sequences of these seven clones contain conserved regions characteristic of catalytic domains of e… Show more

“…After stable transformants were selected with 50 g/ml hygromycin B (Calbiochem), cell lines were cloned by limiting dilution (22). Expression of KFR1 was examined at mRNA and protein levels by Northern hybridization and Western blotting, respectively, as described (16).…”

“…The transformants were then selected under both hygromycin B (50 g/ml) and G418 (50 g/ml) (geneticin, Life Technologies, Inc.). Cell lines were then cloned and examined by genomic Southern hybridization after overnight digestion with PstI or BamHI as described previously (16).…”
