Differential Expression of AtXTH17, AtXTH18, AtXTH19 and AtXTH20 Genes in Arabidopsis Roots. Physiological Roles in Specification in Cell Wall Construction

Abstract: Xyloglucan endotransglucosylase/hydrolases (XTHs) are a class of enzymes that are capable of splitting and reconnecting xyloglucan molecules, and are implicated in the construction and restructuring of the cellulose/xyloglucan framework. Thirty-three members of the XTH gene family are found in the genome of Arabidopsis thaliana, but their roles remain unclear. Here, we describe the tissue-specific and growth stage-dependent expression profiles of promoter::GUS fusion constructs for four Arabidopsis XTH genes, … Show more

“…Eight of these genes are differentially regulated in ise1 and/or ise2 compared with WT (Table 2). XTH19 has been reported as root-specific (36,37), yet it is strongly induced in both mutants. Indeed, XTH19 is the most strongly induced gene in ise1 (50-fold higher than WT levels), and it is also strongly induced in ise2 (15-fold WT levels).…”

“…For example, XTHs are involved in cell elongation by hydrolyzing the cell wall component, xyloglucan, and subsequent loosening of the cell wall (Rose et al, 2002). Among the seven XTH genes that were suppressed in the axr3-1/iaa17-1 mutant, At-XTH17 is expressed in the elongation and differentiating regions of the roots, and At-XTH20 is specifically expressed in the basal mature region of the roots (Vissenberg et al, 2005). Repression of these two XTH genes may be responsible for root growth inhibition in the axr3-1/iaa17-1 mutant.…”

“…Nevertheless, cesa5 mutants have a seed mucilage phenotype (Sullivan et al, 2011), and vegetative plants are indistinguishable from the wild type (Desprez et al, 2007). XTH17 is expressed mostly in roots (Vissenberg et al, 2005;Becnel et al, 2006). Furthermore, our FT-IR and monosaccharide analyses do not support this hypothesis of KAK involvement in cell wall processes, because the results did not show any major differences in cell wall composition or between esk1, beem396b, esk1 kak-7, and esk1 kak-8.…”
