Differential regulation of microtubule-associated protein 1B (MAP1B) in rat CNS and PNS during development

Abstract: MAP1B is a major cytoskeletal protein in growing axons and is strongly regulated during brain development. The present studies compare the expression of MAP1B mRNA, the protein, and its phosphorylated isoform in spinal cord and dorsal root ganglia (DRGs) with brain. In spinal cord and brain, MAP1B mRNA levels were highest in early stages of development, decreased several fold during postnatal development, and remained low in adults. In contrast, there were no significant changes of MAP1B mRNA levels during dev… Show more

“…On the other hand, downregulation of MAP1B in the neonatal brain coincides with the completion of the vigorous extension of neuronal processes and the initiation of active synaptogenesis (28). It is not understood whether the developmentally programmed down-regulation of MAP1B is physiologically important or simply represents a disused phenomenon.…”

“…To visualize axons, we used an antibody against the phosphorylated form of MAP1B-P, which labels axons undergoing different forms of plasticity in the adult: regeneration in PNS 46 and sprouting in the CNS after a periperal nerve lesion. 17,33 Phosphorylated MAP1B-P is also upregulated in axons undergoing sprouting after SCI.…”

“…DRGs neurons were chosen because even at adult stages they survive and regenerate their neurites in vivo and in vitro. In addition, these neurons are special in that they maintain a constitutively high expression of MAP1B in the adult (Ma et al, 1997). Finally, the peripheral axons of the mutant mice used in our study were likely to be affected by the absence of MAP1B because they exhibit a reduced diameter of myelin sheaths and a reduced conductance velocity (Meixner et al, 2000).…”
