Differential requirements for baculovirus late expression factor genes in two cell lines

Abstract: A plasmid library of 18 late expression factor (LEF) genes (LEF library) from the baculovirus Autographa californica nuclear polyhedrosis virus (AcMNPV) supports transient expression from a late viral promoter in the SF-21 cell line, derived from Spodoptera frugiperda. We found, however, that this LEF library was unable to support expression from the same promoter in the TN-368 cell line, derived from Trichoplusia ni, which is also permissive for AcMNPV replication. To identify the additional factor(s) require… Show more

“…HRF-1 (host range factor 1) and HCF-1, which are encoded by LdMNPV and AcMNPV, respectively (Lu & Miller 1995;Thiem et al 1996), preclude global protein synthesis shutdown in a cell-line specific manner, thereby restoring the production of progeny viruses. Ld652Y cells are non-permissive for AcMNPV productive infection and induce global protein synthesis shutdown upon infection with AcMNPV (Thiem et al 1996).…”

“…Other ODV capsid-associated proteins with potential involvement in DNA replication include host cell factor HCF-1 (Braunagel et al, 2003) and viral proliferating cell nuclear antigen (PCNA) (O'Reilly et al, 1989). HCF-1 is required for AcMNPV DNA replication in Trichoplusia ni cell lines (Lu and Miller, 1995a) and it augments the ability of this virus to orally infect T. ni larvae (Lu and Miller, 1996).…”

“…Two other AcMNPV genes, p35 and lef-7, were previously shown to activate transient expression of a late reporter gene in SF-21 cells but have little or no effect in equivalent transient expression assays in TN-368 cells (22). Mutant viruses defective in p35 or lef-7 have also been successfully isolated and propagated in T. ni cells (6,7).…”

“…To examine the role of the AcMNPV ie-2 gene during viral infection, we constructed ie-2 mutant viruses with deletions of different functional regions of IE-2. Since ie-2 is not essential for transient trans activation of the late vp39 promoter in TN-368 cells (22), all ie-2 mutant viruses were constructed by using a cell line derived from T. ni cells. The first of these recombinant viruses, vie2Z, contained an insertion of the E. coli lacZ gene within the ie-2 coding sequences ( Fig.…”
