The sustained emergence of new declared bacterial species makes typing a continuous challenge for microbiologists. Molecular biology techniques have a very significant role in the context of bacterial typing, but they are often very laborious, time consuming, and eventually fail when dealing with very closely related species. Spectroscopic-based techniques appear in some situations as a viable alternative to molecular methods with advantages in terms of analysis time and cost. Infrared and mass spectrometry are among the most exploited techniques in this context: particularly, infrared spectroscopy emerged as a very promising method with multiple reported successful applications. This article presents a systematic review on infrared spectroscopy applications for bacterial typing, highlighting fundamental aspects of infrared spectroscopy, a detailed literature review (covering different taxonomic levels and bacterial species), advantages, and limitations of the technique over molecular biology methods and a comparison with other competing spectroscopic techniques such as MALDI-TOF MS, Raman, and intrinsic fluorescence. Infrared spectroscopy possesses a high potential for bacterial typing at distinct taxonomic levels and worthy of further developments and systematization. The development of databases appears fundamental toward the establishment of infrared spectroscopy as a viable method for bacterial typing.

Mid‐infrared (mid‐IR) spectroscopy (4000–400 cm −1 ) has great potential for determining biochemical properties of microbes that are important to food quality and safety. For example, rapid and precise measurements of pathogenic and spoilage microorganisms present in food are possible, thanks to recent advances in detector technology and multivariate analysis. With mid‐IR spectrometry it is possible to determine compositional properties of bacterial cell membranes, providing useful information about the genotype as well as phenotypical characteristics. In general, the most significant spectral regions for evaluation are Amide I (∼1650 cm −1 ), Amide II (∼1540 cm −1 ), polysaccharide (1200–900 cm −1 ), nucleic acid (∼1240 and ∼1080 cm −1 ), and certain lipid features, such as ω‐cyclic fatty acids (∼2929 cm −1 ). Much research has been conducted to discriminate and quantify the specific microorganisms using this technique since the 1990s. In this chapter, we review recent work focusing on standardizing chemometric‐processing steps, investigations of bacterial biofilms, spore characterization, microbial injury, and bacterial growth. Advantages and disadvantages of the application of IR spectral techniques for microbiological research are also presented.

BACKROUND: Hydrocarbon contamination of groundwater resources has become a major environmental and human health concern in many parts of the world. Microbial degradation of hydrocarbons, through either naturally occurring processes or engineered systems, has been successfully used to reduce concentrations of these pollutants. In order to develop an aerobic bioreactor tailored for this purpose, the present study aims to investigate petroleum contaminated wastewater remediation and toxicity reduction by acclimated microbial consortium enriched in a continuous stirred tank bioreactor (CSTR). Characterization and quantification of refinery wastewater components were performed by gas chromatography mass spectrometry (GC/MS) and Fourier transform infrared (FTIR) spectroscopy. RESULTS:The petroleum hydrocarbons were significantly degraded (97%) by the microbial consortium. After continuous aerobic treatment in the CSTR, the COD effluent and BOD 5effluent average removals were high reaching 97% and 78%, respectively. Also, strong degradation of C10-C35 n-alkanes was observed. The concomitant use of GC/MS and FTIR proved to be a useful complementary tool to assess the impact of treatment strategies on hydrocarbon-contaminated wastewater. In addition, the toxicity of the contaminated wastewater decreased drastically after bioremediation.CONCLUSION: This work shows the ability of this consortium to degrade hydrocarbons and reduce toxicity, which makes them useful candidates for environmental restoration applications at other hydrocarbon-contaminated environments.
