Differentiation of the follicle-associated epithelium in ileal Peyer?s patch and production of 50-nm particles are maintained in B-cell-depleted fetal sheep

Abstract: To evaluate the dependence of the differentiation of the follicle-associated epithelium (FAE) on the presence of follicular B-cells, the FAE of ileal Peyer's patch follicles was examined in B-cell-depleted fetal lambs. The FAE of these rudimentary follicles, which are devoid of lymphocytes, showed normal differentiation, including carbonic anhydrase reactivity and ultrastructural characteristics of transcytosis, extensive interdigitation of the lateral plasma membrane and the shedding of membrane-bounded parti… Show more

“…The early differentiation of epithelial cells into FAE might be dependent on the early presence of B-cells [89]. However, the FAE in ileal PP of B-cell depleted fetal lambs showed a normal differentiation [60]. FAE and M-cells are very important for phagocytosis and transcytosis of external antigens [57].…”

“…Follicular dendritic cells that are reactive with 5' nucleotidase and Mg 2+ dependent adenosine triphosphatase are also seen in the ileal PP as well as in other primary lymphoid follicles in peripheral lymph nodes [43,75]. Depletion of Bcells from fetal lambs by anti IgM antibody injection generated PP follicles composed of clusters of FDC's [60,92]. Ileal PP follicles have also been used to generate a number of clones of mesenchymallyderived stromal cells [38,39].…”

“…This indicates that the signals for M cell differentiation vary between different anatomical compartments. In addition, previous studies of B cell-depleted fetal sheep showed that ileal PPs contain differentiated FAE and reticular cells [20,24]. Therefore, we speculate that development of the FAE in primary lymphoid organs is distinct from that of FAE in secondary lymphoid organs.…”
