Differentiation potential of neural stem cells derived from fetal sheep

Abstract: Neural stem cells (NSCs) are multipotent stem cells that can differentiate into many cell types in vitro. In this study, we isolated and established an NSC line from fetal Ovis aries. Based on the results of immunofluorescence staining, NSCs expressed Nestin, Pax6 and MAP2. Moreover, a reverse transcription–polymerase chain reaction assay was used to biologically characterize the cell line. NSCs were induced to differentiate into neurogenic cells in vitro. They expressed MAP2, glial fibrillary acidic protein (… Show more

“…The PDT was significantly prolonged, cell transparency and viability decreased beginning at passage10 (Fig 3). The 'S' shaped curve obtained shows similarity with the proliferation rate observed by Li et al (2017); Kumar et al (2013) and Agarwal et al(2014).…”

“…In primary cultures, initial adherent colonies were observed within 2 to 3 days (Fig 1A), the cells grew more rapidly and reached 70-80% confluence on 6 to 7 days of culture. The cells were developing into multiple neurites extending from the cell body whose projections intertwined into a network after a week (Fig1B) as illustrated by Li et al (2017). Morphologically similar cells isolated from goat foetal brain were maintained for several passages by Agarwal et al (2014).…”

“…TD = t (lg2 / lgNt—lgN0) (TD and t represent cell doubling time and duration of cell culture, respectively. While, N 0 represent cell number after inoculation and N 1 represent cell number after culturing for t hours [ 15 ]. The Ni was 2000 in each passage and NF was calculated based on number of the harvested cells at 7th day of each passage.…”
