Direct axillary shoot regeneration from the mature seed explant of the hairy vetch (Vicia villosa Roth)

Abstract: The hairy vetch (Vicia villosa Roth) is a climbing, prostrate or trailing legume grown as forage. It fixes atmospheric nitrogen, reduces soil erosion and provides an instant mulch. Multiple axillary shoot regeneration from a mature seed explant (zygotic embryo with two cotyledons) was obtained on MS medium containing 0.05 - 1.6 mg/l TDZ with or without 0.10 mg/l IBA. The frequency (%) of shoot regeneration ranged from 45.83-75.00% with a maximum number of 28.6 shoots per explant on MS medium containing 0.… Show more

“…2b). Our results are in accordance with that of Aasim et al (2011) who achieved direct shoot regeneration from seed explants of Vicia villosa Roth on MS medium supplemented with Thidiazuron and IBA in combination. Ghosh and Sen (1994) also obtained maximum shoot regeneration from shoot tip explants of Asparagus cooperi Baker on MS medium supplemented with BAP in combination with NAA.…”

“…Callus induction started late after 3-4 weeks of culture in this study. Likewise, late callus induction from seed explant has been reported by Aasim et al [25] in hairy vetch and Javed et al [26] in Brassica napus. It was also noted that growth variants in the culture medium were more supportive for shoot induction rather than callus induction.…”

“…The suppressive effects of TDZ on shoot length might be consistent with its high cytokinin activity [37]. On the other hand, addition of IBA helped to overcome the negative effects of cytokinins; a has been reported in hairy vetch [25], fenugreek [38] and narbon vetch [39]. …”

“…Multiple axillary shoot regeneration from mature seed explants has been documented in hairy vetch (Aasim, Şahin Demirbağ, Khawar, Kendir, & Özcan, 2011) and related species of Vicia (Sancak, Mirici, & Özcan, 2000; Selva, Stouffs, & Briquet, 1989). However, no report of root generation from leaf or stem tissue has been published, to our knowledge, at this time.…”
