Disruption and basic functional analysis of six novel ORFs of chromosome XV from Saccharomyces cerevisiae

Abstract: We report the disruption and functional analysis of six open reading frames (ORFs) from chromosome XV, namely YOL155c, YOL154w, YOL119c, YOL118c, YOR301w and YOR306c, in FY1679 and CEN.PK2 backgrounds. We constructed replacement cassettes and cloned each ORF into the pRS416 centromeric plasmid. No obvious phenotype was observed for the corresponding deleted strains with respect to growth, mating or sporulation. YOL155c encodes a protein with a secretion signal and putative GPI-anchor recognition site and is po… Show more

“…The physiological functions and substrate specificities of these proteins were completely unknown. It has been reported previously that the growth of a yol119c yor306c double disruptant in acetate, lactate and pyruvate was similar to that of the parental strain (Lafuente and Gancedo, 1999).…”

“…RgsA (362 amino acids) is similar to ScRgs2p and contains the conserved RGS domain at the N-terminus (Versele et al ., 1999). RgsB (371 amino acids) is similar to ScRax1p in which RGS domain resides at the N-terminus and three putative transmembrane domains are located at the C-terminus (Lafuente and Gancedo, 1999). RgsC (1255 amino acids) is structurally similar to RGS-PX1 (Zheng et al ., 2001), which contains Phox (PX), PX-associated (PXA) and RGS domains.…”

“…In large‐scale screens for mutant phenotypes, qualitative analysis is the rule. Qualitative phenotypic analyses on agar plates have been applied to a large set of yeast mutants within the EUROFAN II project and there have lately been several reports describing analysis of deletion mutants under a limited number of growth conditions (Bilsland et al , 1998; Chan et al , 2000; Hajji et al , 1999; Lafuente and Gancedo, 1999; Rodriguez‐Navarro et al , 1999; Wysocki et al , 1999; Zuniga et al , 1999). However, wider screens encompassing hundreds of chemicals affecting diverse cellular features have also been attempted and, by applying robotics and screens on agar plates, hundreds of mutants have been investigated (Rieger et al , 1999a,b, 1997; Ross‐Macdonald et al , 1999).…”
