Dissection of central clock function in Drosophila through cell-specific CRISPR-mediated clock gene disruption

Abstract: In Drosophila, ~150 neurons expressing molecular clock proteins regulate circadian behavior. Sixteen of these neurons secrete the neuropeptide Pdf and have been called ‘master pacemakers’ because they are essential for circadian rhythms. A subset of Pdf+ neurons (the morning oscillator) regulates morning activity and communicates with other non-Pdf+ neurons, including a subset called the evening oscillator. It has been assumed that the molecular clock in Pdf+ neurons is required for these functions. To test th… Show more

“…However, inactivating 5-HT7 expressing neurons caused food-deprived larvae to avoid GA, similar to the phenotype obtained by inactivating all uPNs. We also specifically removed 5-HT7 from the uPN pathway using a CRISPR/Cas9-based cell type-specific gene knockout system (Schlichting et al, 2019;Delventhal et al, 2019). Without 5-HT7 in the uPNs, food-deprived larvae also avoided GA ( Fig.…”

“…The resulting colonies were sequenced and the correct constructs were inserted into the attP1 landing site on chromosome II (BDSC #8621) by ΦC31mediated recombination (Rainbow Transgenic Flies, Camarillo, CA, USA). Transgenic flies were back-crossed to w 1118 and balanced using BDSC #3703 (Schlichting et al, 2019;Delventhal et al, 2019).…”

“…We will point out how signaling mechanisms may influence circadian clocks in each neuron differently, and how these signals can be integrated to influence neuronal activity. We begin by pointing out that targeted CRISPR-mediated elimination of PER or TIM in the LNvs knocks out the “master pacemaker clock,” yet permits rhythmic behavior in constant darkness ( Delventhal et al, 2019 ).…”

“…14 More recent work suggests, however, that complex interactions of these neurons regulate fly behavior. [15][16][17][18][19][20][21][22] A key feature of circadian clocks is their ability to adjust to the switch from day to night by a process called entrainment. To do so, the central clock can use external stimuli, called zeitgebers, which will allow the animal to predict dawn or dusk (reviewed by Golombek and Rosenstein 23 ).…”
