Distinct proteomic features of two fibrogenic liver cell populations: Hepatic stellate cells and portal myofibroblasts

Abstract: In chronic liver diseases, the accumulation of extracellular matrix leading to fibrosis is caused by myofibroblasts, the origins of which are debatable. We performed a comparative proteomic study to identify markers and gain insight into distinct functions of myofibroblasts derived either from hepatic stellate cells (HSCs) or from portal mesenchymal cells. After isolation from normal liver and culture in similar conditions, myofibroblastic HSCs (MF-HSCs) presented enlarged cytoplasms whereas portal myofibrobla… Show more

“…The PFs are spindle shaped and exhibit biological similarities with activated HSCs; however, they have different genetic profiles and signaling responses (11,12). They are of mesenchymal origins that undergo myofibroblastic differentiation.…”

“…In contrast, the density of CYGB-positive cells (Figure 7d) and CRBP-1-positive cells (Figure 7e) was inversely correlated with the progression of liver fibrosis; the density of CYGB-positive cells was 17.9±1. 29,19.7±1.01, 16.2±0.82, and 13.8±1.06 cells/mm 2 in F1, F2, F3, and F4, respectively, and the density of CRBP-1-positive cells was 9.56 ± 1.24, 14.6 ± 0.77, 12.1 ± 0.83, and 9 ± 0.67 cells/mm 2 in F1, F2, F3, and F4, respectively.…”

“…29 In particular, the microfilaments of myofibroblasts contain a-SMA that is the actin isoform typical of smooth muscle cells located in the vessel wall 30 and has become the most reliable marker for myofibroblastic cells. 31,32 Myofibroblasts are additionally positive for Thy-1.…”

“…Various markers have been used to distinguish portal fibroblasts from hepatic stellate cells, including fibulin-2, IL-6, elastin, nucleoside triphosphate diphosphohydrolase-2 (NTPDase2), cofilin-1 and neuronal proteins such as synaptophysin 2,6,10,11 . We have found that elastin is a good marker for portal fibroblasts, even after myofibroblastic differentiation, while NTPDase2 is lost after portal fibroblasts have undergone culture after several days 9 .…”
