Distinction between Human Cytochrome P450 (CYP) Isoforms and Identification of New Phosphorylation Sites by Mass Spectrometry

Abstract: In mammals, Cytochrome P450 (CYP) enzymes are bound to membranes of the endoplasmic reticulum and mitochondria, where they are responsible for the oxidative metabolism of many xenobiotics as well as organic endogenous compounds. In humans, 57 isoforms were identified which are classified based on sequence homology. In the present work, we demonstrate the performance of a mass spectrometry-based strategy to simultaneously detect and differentiate distinct human Cytochrome P450 (CYP) isoforms including the highl… Show more

“…Using LC-MS/MS analyses, we have identified several CYP2E1 residues, in addition to the previously identified Ser 129 (28,(51)(52)(53) specifically phosphorylated by PKA or PKC in the native enzyme. Furthermore, we document that this phosphorylation is further enhanced after CYP2E1 structural inactivation by cumene hydroperoxide (CuOOH).…”

“…Human P450 3A4 is found mainly in the liver and accounts for the metabolism of about 50% of all xenobiotics [70]. Highly promiscuous by nature due to its large and flexible active site, substrate prediction with this enzyme has been very challenging.…”
