Distribution of aldehyde dehydrogenase 1‐positive stem cells in benign mammary tissue from women with and without breast cancer

Abstract: This study provides further evidence for the stem cell character of ALDH1+ cells, here in benign breast tissue of cancer and non-cancer patients throughout non-lactating adult life, and contributes evidence of benign stromal ALDH1+ cells. The distribution of ductular ALDH1+ stem cells appears to be influenced by hormonal status.

“…Although contradictory results have been reported for ER expression in CSCs [20], our results for DCIS and IDCs [5] strongly suggest that most CSCs do not express ER or PgR. In addition, we could demonstrate that ALDH1-positive epithelial cells did not overlap with ER- or Ki67-positive epithelial cells in normal breast tissues, as previously reported [21]. These results seem to indicate that CSCs in both DCIS and IDCs as well as stem cells in the normal breast gland have the same characteristics, i.e., most of them are in the G0 phase and unlikely to express ER and PgR.…”

“…ALDH1 expression has been detected in benign breast tissue as well as in tumor tissue [31,32]. In tumor tissues, ALDH1 was expressed in both tumor and stromal cells.…”

“…Interestingly, stromal expression has been reported to be present in all cases of benign breast tissue and also to be more common in benign than malignant breast lesions. This may suggest that strong stromal ALDH1A1 expression more resembles normal breast tissue, while the loss of ALDH1A1 represents a more tumour-promoting milieu 22 42. ALDH1A1 expression has been reported in both spindle-shaped cells and round stromal cells, the latter presumed to be mainly lymphocytes 22.…”

ALDH1A1 expression has been reported in both spindle-shaped cells and round stromal cells, the latter presumed to be mainly lymphocytes 22. We therefore chose to evaluate the spindle-shaped cells, presumed to be fibroblasts, as the immune response and number of infiltrating lymphocytes are known prognostic factors.…”
