Distribution of genotype/subtype and mutational spectra of the surface gene of hepatitis B virus circulating in Hanoi, Vietnam

Abstract: In order to ascertain the molecular epidemiological features and mutational spectra of hepatitis B virus (HBV) in Hanoi, Vietnam, direct sequencing of the 219-nucleotide fragment of the surface (S) gene of HBV from the sera of 40 patients mostly with chronic hepatitis were carried out. The samples were classified into genotypes by phylogenetic and genotype-specific analysis, and subtypes by the deduced amino acid sequences. The results showed that genotype B with ayw1 was predominant genotype/subtype (63%), fo… Show more

“…Unfortunately, the vaccination status of the mother from whom the HBV strain was obtained is unknown. Studies carried out in Morocco and in Vietnam reported mutations in these same positions: L109Q and G130A/N in unvaccinated patients [ 52 , 61 ].…”

“…Because it replicates through an RNA intermediate synthesized by reverse transcriptase, mutant viral genomes[5972] and quasi-species[7177] are generated. This results in the production of viral mutants during naturally occurring infections.…”

“…[5976808187] Some mutations involve amino acid insertions into the a determinant[63788889] or the creation of stop codons. [7184] Other mutations eliminate the glycosylation site at residue 146[85] or create potential new glycosylation sites in the first loop of the MHR. [9091] However, not all mutations in the MHR lead to escape mutants.…”

“…Limited research has been conducted on molecular characterization and determination of virulence associated properties of HBV in Vietnam. Representative data on genotype and subgenotype analysis using whole genome sequence (WGS) [ 14 16 ], as well as information on the prevalence of virulence associated mutations in different genes, recombination and drug resistant mutations in treatment naïve patients are limited.…”
