Distribution of IgM and IgG antibodies to oxidized LDL in immune complexes isolated from patients with type 1 diabetes and its relationship with nephropathy

Abstract: Modified lipoproteins are immunogenic and play a key pathogenic role in vascular disease. Antibodies to oxidized LDL (oxLDL) are mostly of the proinflammatory IgG 1 and IgG 3 isotypes. We measured IgG and IgM oxLDL antibodies in immune complexes (IC) isolated from 36 patients with type 1 diabetes using a nested case-control design. IgG antibodies predominated over IgM antibodies by an 8:1 ratio. IgG antibody concentrations were higher in the nephropathy cases compared to controls (p=0.09), but no significant d… Show more

“…The circulating IC were precipitated from serum samples with 4% polyethylene glycol, resuspended, and fractionated in immobilized protein G columns. IgG and IgM oxLDL antibodies were assayed in the eluate and washout fractions of the Protein G column, respectively [83]. This approach has the advantage of measuring the specific antibodies of true significance, i.e., those able to form stable IC.…”

“…It has been reported that lymphocytes from human atherosclerotic plaques recognize oxidized LDL (ox-LDL) in vitro [9]. The induction of autoantibodies against ox-LDL has also been reported in several other studies [10,11,12,13,14,15,16,17]. There are several epitopes of ox-LDL that induce autoimmune responses in humans.…”

“…The oxLDL particles bound to antibodies are taken up from the circulation via Fcγ II receptors present in cells from the mononuclear phagocytic system contributing to the inflammation in the vascular wall [11][12][13][14]. Involvement of these receptors in atherosclerosis has been recently demonstrated and decreased expression of FcγRII was found in patients with cardiovascular diseases [15].…”
