DiSUMO-like DSUL is required for nuclei positioning, cell specification and viability during female gametophyte maturation in maize

Abstract: Reversible post-translational modification of numerous proteins by small ubiquitin-related modifiers (SUMOs) represents a major regulatory process in various eukaryotic cellular and developmental processes. To study the role of sumoylation during female gametophyte (FG) development in maize, we identified Zea mays genes encoding SUMO (ZmSUMO1a and ZmSUMO1b) and a diSUMO-like protein called ZmDSUL that contains two head-to-tail SUMO-like domains. Whereas ZmSUMO1a and ZmSUMO1b are almost ubiquitously expressed, … Show more

“…In FG mutants with abnormally positioned or extra nuclei in maize (Evans, 2007;Srilunchang et al, 2010) and Arabidopsis (Gross-Hardt et al, 2007;Johnston et al, 2010;Moll et al, 2008;Pagnussat et al, 2007), the nuclei acquire their fate according to their spatial positioning along the micropylar-chalazal axis, suggesting that cell specification in the FG is driven by positional information. This hypothesis was strengthened by a microscopy study showing a graded activity of the auxin-sensitive reporter DR5 (Ottenschläger et al, 2003) along the micropylar-chalazal axis of the FG (Pagnussat et al, 2009).…”

“…Until recently, there were only a few specific promoters defined in maize (Cao et al, 2007, Sattarzadeh et al, 2009, Srilunchang et al, 2010, Zhang et al, 2009. We generated a set of promoter-FP direct fusion marker lines, which can be used for FACS experiments.…”

“…The gene represented 17 by the EST sequence EPS87 (TaDSUL) codes for a protein with small ubiquitin-like modifier 18 (SUMO) and ubiquitin-like domains (di-SUMO-like or DSUL, gene bank accession: 19 ACL50300.1). Recently, the maize homologue of this protein has been characterized in detail 20 by Srilunchang et al (Srilunchang et al 2010) who specified that it is required for nuclei 21 positioning, cell specification and viability during female gametophyte maturation. 22…”

“…The maize homologue of TaDSUL, a 1 protein may be involved in the post-translational regulation of other proteins by sumoylation 2 during female gametophyte maturation (Srilunchang et al 2010). This gene was reported to 3 be exclusively expressed in the micropylar region of the immature female gametophyte, while 4 after cellularization its expression was restricted to the egg cell and the zygote (Srilunchang et 5 al.…”
