Diterpenoids and a Diarylheptanoid from Hedychium coronarium with Significant Anti‐Angiogenic and Cytotoxic Activities

Abstract: Two new labdane diterpenoids, namely hedycoronals A and B (1 and 2, resp.), were isolated from the rhizomes of Hedychium coronarium, together with eight known diterpenoids, 4-11, and a known diarylheptanoid, 3. The structures of 1 and 2 were established by detailed interpretation of their 1D- and 2D-NMR spectra and HR-ESI-MS data. Inhibitory activities against human umbilical vein endothelial cells (HUMECs) proliferation and cytotoxic activities against four cancer cell lines were assessed for all the isolates… Show more

“…Suresh et al [90] showed the presence of two new labdane diterpenes along with coronarin D and C, coronarin D methyl ether, cryptomeridiol, hedychenone, pacovatinin, 6-oxo-7,11,13-labdatriene-16,15-olide, 4-hydroxy-3methoxy cinnamaldehyde and 4-hydroxy-3-methoxy ethyl cinnamate from the plant with cytotoxic effects on A-549 (lung cancer), SK-N-SH (human neuroblastoma), MCF-7 (breast cancer) and HeLa (cervical cancer) cell lines. Similar work carried out by Zhan and colleagues showed positive cytotoxic and antiinflammatory effects of common isolates upon B16, HT-29, HeLa and HepG2 cell lines [91] . During inflammatory reciprocation, activation of the nuclear transcription factor-kB (NF-kB) is an essential step [92] .…”

“…The HepG2 antiproliferative activity exhibited by compound 1 could be related to ARE induction activity, since the ARE-luciferase plasmids transfected cells are constructed from the HepG2 cell line. The low cytoxicity of compound 2 in cancer cells MCF-7, A549 and SK-N-SH (Suresh et al, 2010), and Hela and HUMEC has been also reported (Zhan et al, 2012).…”

“…A broad spectrum of biological activities has been described for labdane diterpenes, including antibacterial, antiviral, antiinflammatory and anti-tumor activities (Ghosh and Karin, 2002;Pezzuto et al, 2005;Maiti et al, 2007;Girón et al, 2008;Hegazy et al, 2008;Kunnumakkara et al, 2008;Suresh et al, 2010;Zhan et al, 2012). Remarkable inhibitory activity of nitric oxide (NO) production in LPS-activated mouse peritoneal macrophages has been reported for coronarin D and hedychilactone A, labdane diterpenes isolated from the rizhomes of H. coronarium Kiem et al, 2012).…”

“…Cytotoxic effect on cancer cells is the prime concern of any cancer therapy, which can be attained by one or several pathways (Chemotherapy Principles, ). Hedycoronals A and B isolated from Hedychium coronarium were found to exert moderate‐to‐potent cytotoxic effects on human umbilical vein endothelial cells (Zhan et al , ). On the other hand, the dehydroabietyl skeleton diterpenes exhibited cytotoxicity within half‐minimal inhibitory concentration (IC 50 ) values 0.75 to 10.65 μM in HCC (SMMC‐7721), lung cancer (A‐549), glioma (C‐6) and breast carcinoma (MCF‐7) cells (Rao et al , ).…”
