Diverse preparation methods for small intestinal submucosa (SIS): Decellularization, components, and structure

Abstract: The native extracellular matrix (ECM) biomaterial derived from small intestinal submucosa (SIS) is widely applied in tissue engineering for tissue repair and regeneration. SIS ECM is obtained through physical and chemical methods to remove the intrinsic cells, which would otherwise cause adverse immune reactions when the SIS ECM is implanted into the host body. Several research teams have reported diverse SIS decellularization methods. However, there was no consensus on the criteria to be used for the decellul… Show more

“…The natural biomaterials have good biochemical activity and biological compatibility. However, in the process of handling natural materials, such as the decellularized tissues, the normal 3D structure is changed or some biological factors are lost [ 7 ]. Furthermore, the requirements of material characteristics vary with different kinds of tissue construction.…”

“…And bioabsorbable collagen membrane is the most frequently used barrier membrane in GBR treatments [6,7]. e extracellular matrix (ECM), which retains collagen and various signaling molecules containing glycosaminoglycans, glycoproteins, and abundant growth factors [8], has been used in the GBR technique for the purpose of building a tissue engineering scaffold [9][10][11][12]. Porcine-derived small intestinal submucosa (SIS) is an acellular, naturally derived ECM material from the submucosal layer of porcine intestinal tissue [13,14].…”

“…Although still undocumented as an efficient decellularized agent, trypsin can be combined with physical, acid-, base-, or detergent-based protocols [80][81][82]. The action of trypsin is achieved by immersion of the tissue in an isotonic solution containing from 0.25% to 0.1% w/v of enzyme, separately from the other treatments of the protocol.…”
