Diversity and dynamics of plant genome size: An example of polysomaty from a cytogenetic study of Tahitian vanilla (Vanilla ×tahitensis, Orchidaceae)

Abstract: Genome irregularities may be transmitted not only during vegetative propagation but also by sexual reproduction in Vanilla. However, there must be a complex regulation of genome size and organization between the aneuploidy in somatic tissues and subsequently euploid gametic tissue. This is a novel example of polysomaty with developmentally regulated partial endoreplication.

“…The number of linkage groups was higher than expected according to the haploid number of chromosomes found in diploid tahitian vanilla ( n = 15; Lepers‐Andrzejewski et al, 2011). Furthermore two linkage groups were of less than 20 cM, indicating a lack of mapping markers and marker sparse regions.…”

“…Three ploidy levels, ranging from diploid to tetraploid, have been observed among accessions of tahitian vanilla (Lepers‐Andrzejewski et al, 2011). Tahiti, which is diploid, is the morphotype most common in French Polynesia.…”

“…[26] Solanum lycopersicum L. "Montfavet 63/5 (2C=1.99 pg) was used as internal standard. [27] In order to remove large tissue debris and fragments, the nuclei suspension was filtered through nylon mesh, with 50 μm size of pore and stained with 50 μg/mL propidium iodide (Sigma Chemical Co. St. Louis, USA). At least 5 individuals with 5000 to 10000 analyzed nuclei were measured to obtain the mean DNA content.…”
