Diversity of Microbial Communities of Pinus sylvestris var. mongolica at Spatial Scale

Abstract: Soil microorganisms play an indispensable role in the forest ecosystem. It is necessary to study the soil microorganisms in Pinus sylvestris var. mongolica, which is one of the afforestation species widely planted in the northern sandy region of China. We collected soil samples of P. sylvestris at large spatial scales and analyzed bacterial and fungal community composition differences using high-throughput sequencing techniques. The results showed that: (1) the richness index of different sandy lands was signi… Show more

“…The diversity indices and community structure of EcM fungi changed with stand aging in the Mu Us Desert, but the effect of forest age was not significant. This was consistent with previous studies that the diversity of the fungal community was not correlated with the aging stand (Wang et al, 2020). As expected, the EcM fungal community was dominated by Basidiomycota rather than Ascomycetes, which is consistent with previous results (Geml et al, 2017).…”

“…Microbial diversity improves the productivity and stability of the ecosystem [52,53,54]. In this study, the soil with press mud had higher alpha diversity than the soil without press mud in both sites after heavy deposition of organic matter (Table 1).…”

“…Furthermore, bacterial alpha diversity is notably influenced by soil chemical properties, including soil organic carbon (SOC), total nitrogen (TN), and available phosphorus (AP). Our previous study demonstrated that at broader spatial scales, temperature and precipitation primarily drive variations in soil microbial communities within Pinus sylvestris forest [ 28 ]. Notably, bacterial community composition is predominantly influenced by chemical properties, including total nitrogen, soil organic carbon, and pH levels.…”

“…The amplification of the V3–V4 region of the bacterial 16S rRNA gene employed primers 338F (ACTCCTACGGGAGGCAGCA) and 806R (GGACTACHVGGGTWTCTAAT) in a 25 μL system. Purification and quantification procedures adhered to the protocol outlined by Wang et al [ 28 ]. For purification and recovery, magnetic beads (Vazyme VAHTSTM DNA Clean Beads) were added at 0.8 times the volume of the PCR product.…”
