Diversity of operation in ATP-dependent chromatin remodelers

Abstract: Chromatin is actively restructured by a group of proteins that belong to the family of ATP-dependent DNA translocases. These chromatin remodelers can assemble, relocate or remove nucleosomes, the fundamental building blocks of chromatin. The family of ATP-dependent chromatin remodelers has many properties in common, but there are also important differences that may account for their varying roles in the cell. Some of the important characteristics of these complexes have begun to be revealed such as their inter… Show more

“…Indeed, remodeller target site on DNA could be located differently with respect to the histone octamer. While the translocase domain of yIsw2 interacts with the external face of nucleosomal DNA, the hSnf2 binds with high affinity the DNA gyre towards the octamer (Hota and Bartholomew 2011). Interestingly, structural studies of yeast Isw1a suggest that ISWI remodellers can establish additional contacts with the DNA, allowing them to bind di-nucleosomes and to sense the length of the linker DNA (Yamada et al 2011).…”

“…2) (Boyer et al 2004;Grune et al 2003b;Hota and Bartholomew 2011). Over the past few years, in vivo and in vitro studies have led to a broadly accepted model in which the HSS domain plays an integral part during the remodelling reaction (Boyer et al 2004;Grune et al 2003a;Boyer et al 2002).…”

“…To assist RNAPII passage, chromatin remodelers and histone chaperones aid in removal of the nucleosome blockade. Chromatin remodelers unwind DNA from nucleosomes in an ATP-dependent manner [16] whereas histone chaperones are enzymes that facilitate exchange of histone variants during transcription in a histone-dependent, ATP-independent manner [17]. In addition to histone modifications and chromatin remodeling, replacement of canonical core histones with specialized variants assists in regulation of the open or closed states of chromatin [18] [19].…”
