Does Addition of Propolis to Glass Ionomer Cement Alter its Physicomechanical Properties? An in Vitro Study

Plants may contain beneficial or potentially dangerous substances to humans. This study aimed to prepare and evaluate a new drug delivery system based on a glass-ionomer-Brazilian pepper extract composite, to check for its activity against pathogenic microorganisms of the oral cavity, along with its in vitro biocompatibility. The ethanolic Brazilian pepper extract (BPE), the glass-ionomer cement (GIC) and the composite GIC-BPE were characterized by scanning electron microscopy, attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR), and thermal analysis. The BPE compounds were identified by UPLC–QTOF–MS/MS. The release profile of flavonoids and the mechanical properties of the GIC-BPE composite were assessed. The flavonoids were released through a linear mechanism governing the diffusion for the first 48 h, as evidenced by the Mt/M∞ relatively to $$\sqrt t$$ t , at a diffusion coefficient of 1.406 × 10–6 cm2 s−1. The ATR-FTIR analysis indicated that a chemical bond between the GIC and BPE components may have occurred, but the compressive strength of GIC-BPE does not differ significantly from that of this glass-ionomer. The GIC-BPE sample revealed an ample bacterial activity at non-cytotoxic concentrations for the human fibroblast MRC-5 cells. These results suggest that the prepared composite may represent an alternative agent for endodontic treatment.

ASSESSMENT OF MICROLEAKAGE AND MICROHARDNESS OF GLASS IONOMER CEMENT CONTAINING PROPOLIS: In Vitro Study

INTRODUCTION:Propolis is a natural product collected by bees. Contemporary dentistry has recently focused on it, due to its antimicrobial properties. OBJECTIVES: To evaluate and compare the microleakage and microhardness of GIC containing EEP at 2 different concentrations to the conventional GIC. MATERIALS AND METHODS: Two tests were assessed 1. Microleakage test :where 24 exfoliated primary teeth were divided randomly into three groups according to the restorative material used after standardized class V cavity preparation: (Group1: Conventional GIC, Group2: GIC containing 25% EEP, and Group 3: GIC containing 50% EEP), teeth were immersed in 2% methylene blue solution for 24 hours, sectioned and examined under stereomicroscope. 2. Microhardness test: 8 specimens from each group (GIC, GIC 25% EEP, and GIC 50% EEP) were prepared using a mold of six mm in diameter and two mm in thickness. The hardness of the specimens was measured using Vickers Micro-hardness Tester. RESULTS: There was no statistically significant difference in the microleakage scores between the three groups (p=0.34).The Microhardness values of the present study showed that GIC with 25% EEP showed significantly higher values than both the conventional GIC and GIC with 50% EEP (p<0.0001). There was no significant difference in the microhardness between the conventional GIC or GIC with 50% EEP (p>0.05) CONCLUSION: The addition of EEP to GIC in either (25%, 50%) did not affect the microleakage of GIC. Addition of 25% EEP increased the microhardness of the GIC while the addition of 50% EEP did not affect the microhardness of GIC.

Background: Dental caries is a major concern to the dentist. Many bacterial subspecies are associated with caries, but Streptococcus mutans is still the most important bacterium in the initiation and progress of this disease. the direction toward natural products for medicinal purposes has gained much attention, as these natural products have proven to be effective with less toxic side effects. Studies have also shown other important properties in propolis such as antibacterial, antifungal, antiviral, anti-inflammatory, anesthetic, and ability to promote healing. Objective: To study the effect of propolis ethanolic extract (EEP) on enhancing the antimicrobial activity of glass ionomer cement (GIC). Methods: Two EEP 25% & 50% concentrations were evaluated for antimicrobial activity, in combination with GIC using agar Disk Diffusion test and broth microdilution test. Results: The diameter of the inhibition zone increased with 25% EEP & 50% EEP over the control by 14.4% and 19.6%, respectively. While increasing the concentration from 25% to 50% resulted in only 5% increase in the inhibition zone. MIC calculation for the three groups revealed, the combination of GIC and EEP has reduced the mic against Streptococcus mutans by two folds. Conclusion: The addition of propolis in different concentrations to GIC increased the antibacterial effect. 25% EEP gave the best antibacterial action with the lowest concentration.
