Does CYP1A1 gene polymorphism affect cell damage biomarkers and ageing?

Abstract: This study aimed to investigate the effects of cytochrome P450 1A1 (CYP1A1) gene polymorphism on early ageing due to occupational exposure to mutagens. The project was conducted on buccal epithelial cells collected from mechanical-workshop workers and controls. Epithelial cells were obtained from the inner part of the cheeks. Techniques including comet assay, micronuclei test, realtime polymerase chain reaction, and restriction fragment length polymorphism-polymerase chain reaction were used to assess DNA dama… Show more

“…In humans, since the CYP1A1 is an extrahepatic protein, it is considered to assume a secondary function in the removal of medications in vivo , and consequently, the polymorphism of the CYP1A1 gene may have a minor impact on their metabolic clearance. CYP1A1 assume essential tasks in the bio-activation of a collection of cancer-causing polycyclic aromatic hydrocarbons [PAHs; e.g ., benzo[a]pyrene (B[a]P)], aromatic amines and amides, and mycotoxins ascertained in certain grains, e.g ., aflatoxin B1[10,38-41]. CYP1A1 is also committed in the metabolism of endogenous compounds such as the pineal hormone melatonin and 17 β-estradiol and estrone[42-44].…”

“…DNA damage effects of the used substances were confirmed in mechanical workshops workers, but with no confirmation of the influence of GSTP1 [66] or CYP1A1 [69] gene polymorphism on DNA damage, considering the comet assay performed on buccal cells. Conversely, workers with the wild genotype for CYP2E1 showed statistically significant higher comet tail length at the occupational exposure, while the mutated genotype did not have influence on this biomarker [67].…”

“…The comet assay in buccal cells has been used to evaluate DNA damage induced by different materials such as mouthrinses [50], metals released from orthodontic appliances [51][52][53][54][55][56][57][58][59], ionizing radiation [60], as well as assessment of DNA damage, and its modulation by life-style, dietary, genetic and healthy factors [61][62][63][64][65][66][67][68][69][70][71][72][73][74], occupational exposure [66][67][68][69][75][76][77][78][79][80][81][82], and environmental exposure [83][84][85][86]. Different procedures have been used in collecting and processing the samples that are presented and discussed in Rojas et al [33].…”

“…The increased cost and time requisite for screening the effect of mutation by conventional laboratory-based experimentations is impossible due to the presence of enormous numbers of SNPs. However, information on mutated gene function is essentially required, and particularly on missense mutations causing hereditary diseases (Eshkoor et al, 2014). The understanding of the relationships between protein structure and function is also a prerequisite.…”
