Double-Blind Evaluation of a Miconazole – Benzoyl Peroxide Combination for the Topical Treatment of Acne Vulgaris

Abstract: The results of a randomized double-blind study show that benzoyl peroxide 5% combined with miconazole nitrate 2% in a cream formulation brought about a good to excellent result in 88% of 51 patients with moderate to severe acne, compared with benzoyl peroxide 5% alone which gave improvement in 41 % of 51 comparable patients. The difference is highly significant. Moreover, the frequency of the well-known irritating and drying effect of benzoyl peroxide was shown to be reduced from 10 cases in the benzoyl peroxi… Show more

“…Although the synergistic activity between [Degreef and Vanden Bussche, 1982] and hence remains to be evaluated in a clinical trial such as the one described in this paper, we infer that the pos tulated synergism could be achieved by ap plying both drugs separately and state, in case that synergistic effects should not be confirmed by other groups, that both thera peutic regimens were equally effective. This was to be expected from the dynamics of per cutaneous penetration inasmuch as the available data can apply to the various bases under study.…”

“…The global interpretation of the integrated information obtained from this dual phase trial is suggestive of a direct effect of miconazole nitrate on the inflammatory process of acne. This medication is active against bacteria 11 and has shown clinical efficacy in treating acne on a long‐term basis 8,14 . The mechanisms leading to the presently reported effect and to the long‐term effect on acne severity are probably different.…”

“…The medicated paste contained miconazole nitrate, which is a time‐honored compound active against many fungi including Malassezia yeasts, and against many gram‐positive bacteria including Staphylococcus spp. and P. acnes (24, 25). All these microorganisms may be involved in acne and acne‐related disorders (2, 26, 27).…”
