Dual Expression of the Salmonella Effector SrfJ in Mammalian Cells and Plants

Abstract: SrfJ is an effector of the Salmonella pathogenicity island 2-encoded type III secretion system. Salmonella enterica serovar Typhimurium expresses srfJ under two disparate sets of conditions: media with low Mg2+ and low pH, imitating intravacuolar conditions, and media with myo-inositol (MI), a carbohydrate that can be used by Salmonella as sole carbon source. We investigated the molecular basis for this dual regulation. Here, we provide evidence for the existence of two distinct promoters that control the expr… Show more

“…Salmonella infection increases the HexCer/Cer ratio of murine macrophages in a SrfJ-dependent manner (Figure 5). The effect of SrfJ in this context is not related to differential intracellular survival, since our previous results showed that the srfJ mutant has no significant defect in intracellular proliferation compared to the wild-type strain [20]. In addition to Cer and HexCer, the abundance of many other lipids is altered by the presence of SrfJ, which could be due either to indirect effects or to other unknown activities of this effector.…”

“…SrfJ is a T3SS2 effector the synthesis of which is positively regulated by SsrB [16], the main regulator of SPI2 [17], and negatively by IolR [18], the regulator of the myo-inositol utilization island [19]. This dual regulation is due to the existence of two distinct promoters that control the expression of the gene srfJ [20]. A proximal promoter, PsrfJ, responds to intravacuolar signals and is positively regulated by SsrB and PhoP and negatively regulated by RcsB.…”

“…SrfJ is a specific effector located in SPI-2 encoded secretion [57]. Studies by Julia et al have provided evidence for downregulation of expression of SrfJ leading to reduced proliferation inside host macrophages [58]. Hence, attempts to analyze the specific binding affinity of oxyresveratrol to residues on SrfJ modelled crystal structure of Salmonella enterica strain used in the study was performed.…”
