“…pMT36 (P hCMV*-1 -PIT), engineered for P hCMV*-1 -driven PIT 17 expression, was constructed by elimination of the NotI fragment from pMF125 (ref. 28). pMT100 (P hCMV*-1 -Ub V76 -GFP), enabling the tetracycline-responsive expression of Ub V76 -GFP, was designed by inserting Ub V76 -GFP, excised from pcDNA3.1-Ub V76 -GFP by NheI/NotI, into the corresponding sites of pMF111 (ref.…”

“…168 Another development pertinent to autoimmune gene therapy is the construction of vectors from which two transgenes can be expressed independently under the control of the tetracycline or streptogramin regulatory systems. 169 These vectors would permit two genes with different therapeutic targets to be expressed in a regulated manner and at a stage during treatment that achieves optimal effect, for example, expression of cytokines with anti-inflammatory and tissue repair activity in the treatment of arthritis.…”

“…In combination with the rtTA system, they should allow inducible gene knockout and rescue via tamoxifeninduced gene recombination and dox-dependent reversible transgene expression (figure 2c). The association of various inducible recombination-expression systems is a promising means for targeting-expressing several genes and fluorescent markers simultaneously (Baron et al 1999;Moser et al 2001) to further advance research on LTP.…”
